Folate (Vitamin B9) is required for one-carbon metabolism and methylation reactions throughout the body. 5-MTHF (5-methyltetrahydrofolate) is the biologically active form that does not require conversion by the MTHFR enzyme, making it preferable for individuals with common MTHFR gene variants.
Clinical Summary
Clinically preferred over synthetic folic acid for individuals with MTHFR polymorphisms, since it bypasses the rate-limiting conversion step.
Dosage Guidelines
400mcg to 800mcg (DFE) daily for general supplementation.
Safety & Side Effects
Generally very safe; high-dose synthetic folic acid (not 5-MTHF) has been associated with masking B12 deficiency symptoms.
Known Interactions
May interact with methotrexate and certain anti-seizure medications; consult a physician if on these drugs.
